The Staining Variations on The Kidney and Liver Tissue Sections with Periodic Acid Schiff and Lemon Juice Solution

Abstract

Periodic acid–Schiff (PAS) is a staining method used to detect polysaccharides such as glycogen, and mucosubstances such as glycoproteins, glycolipids and mucins in tissues. To establish the staining variations between tissue sections stained with Periodic Acid Schiff’s method and that of tissue section stained with Lemon Schiff’s staining method. Tissue samples were gotten from the liver and kidney of adult Wistar rats. The tissues obtained were histological processed and sectioned with a rotary microtome. Lemon orange was obtained from a local market in Anyigba, Kogi State. The juice was extracted locally and used to oxidize tissue section obtained from the liver and kidney from adult Wistar rats before staining the tissue section with Schiff reagent and then counterstaining with Cole’s haematoxylin. The control tissue sections were correspondingly stained with Periodic Acid Schiff’s stain for the demonstration of kidney and liver tissues. Liver and kidney tissue sections oxidized with lemon juice extracts stained more intensely with Schiff’s reagents when compared with the other liver and kidney tissue sections oxidized with Periodic Acid and then stained with Schiff’s reagent. Considering the result generally, lemon juice Schiff reagent is a good substitute for Periodic Acid Schiff reagent.
