Comparison of The Staining Reaction of Lime Juice Solution with Periodic Acid Schiff Reagent on Liver and Brain Tissues

The aim of this study was to compare the histochemical reaction of Periodic Acid Schiff with the histochemical reaction of lime juice Schiff on Liver and Brain tissue section of Wistar rats. Four (4) sections each were obtained from a processed Brain tissue and processed Liver tissue. The limes juice was extracted and the various pH was measured with different timing. The control tissue sections were stained with Periodic Acid Schiff technique while the test tissue sections were stained with the limes extraction obtained in a lime Schiff technique. The results show that both Periodic Acid Schiff and lime juice stained Brain and Liver sections with a similar colour (purple-magenta) and they clearly show the specific cellular features of Brain and Liver tissue. Lime Schiff technique has stained the liver and brain of the Wister rats clearly the same way as periodic acid Schiff. This finding of this study indicates that Lime Schiff technique can possibly be used as a more cost effective and readily available alternative for the demonstration of carbohydrates in Brain and Liver sections particularly in developing countries.
